The Panda II has 4 UART, 2 CAN, 1 I2C, and other things. Now, I can connect to one of the UART’s via the e-block, but how do I get to the other three? The same would apply to the CAN, and the I2C, how and where are the connections?

You need to do some wiring. Please see the FEZ Panda II brochure for pinout.
